Highway A1 FIRENZE-ROMA direction Firenze. Exit BETTOLLE/SINALUNGA (before Florence). Proceed to Siena for about 60 km. In Siena, follow the indicatons for "centro-stazione". After the railway station you will see the indication for Castellina in Chianti (20 km north from Siena). When arrived in town ask at Palazzo Squarcialupi how to reach La Castellina.
